design_verification_primers

design_verification_primers is a skill for Claude Code from GGbond-bo/MemOmics-Agent. It costs 45 tokens per session (1,375 once invoked), scanned A, original, MIT.

A DNA-primer design tool for checking a selected region of a plasmid by Sanger sequencing, a method for reading a short DNA sequence. It reuses existing primers when possible and designs more when coverage is incomplete.

In plain words
What is it for?
Use it to plan verification primers for a plasmid region, including circular plasmids, before Sanger sequencing.
Why use it?
It reduces the manual work of choosing primers that cover the region and meet basic length, GC-content, and temperature limits.

Token cost

What it costs to keep this loaded

Counted locally with the o200k_base tokenizer, which is exact for GPT models; Claude uses its own tokenizer and its counts differ. Treat this as one consistent yardstick across the catalogue rather than a bill. Prices are per million input tokens.

ModelPer sessionOnce invoked
Fable 5.1 $0.00045 $0.01375
Opus 5 $0.00023 $0.00687
Sonnet 5 $0.00009 $0.00275
Haiku 4.5 $0.00005 $0.00137

Security

Grade A, and why

design_verification_primers scanned grade A with 0 findings against 26 rules in 11 categories — prompt injection, anti-refusal, data exfiltration, privilege escalation, supply chain, agent snooping, system-prompt leakage, SSRF and excessive agency — measured 9d ago.

The scan reads SKILL.md. This mod also ships 1 executable file (scripts/run.py), listed below but not scanned — reading those needs a real analyzer, not pattern matching.

A static scan of the body, not an audit. Every finding is printed with the line that produced it so you can judge whether it matters here. A mod is markdown that instructs an agent; that is exactly why what it instructs is worth reading.

Nothing flagged

None of the 26 patterns this scan looks for appear in this file: no shell pipes, no recursive deletes, no credential paths, no hidden text, no instruction-override or anti-refusal phrasing, no agent-config snooping. That is not a guarantee, it is the absence of the things that are checkable.

Design Verification Primers

Design Sanger sequencing primers to verify a specific region in a plasmid. First tries to use primers from an existing primer pool. If they cannot fully cover the region, designs additional primers as needed.

When to Use

When you need design verification primers analysis

Parameters

Parameter Default Notes
plasmid_sequence [Required] The complete plasmid sequence (str)
target_region [Required] Start and end positions to verify (0-based indexing) (Tuple[int, int])
existing_primers [Optional] List of existing primers with their sequences and optional names
is_circular [Optional] Whether the plasmid is circular (default: True)
coverage_length [Optional] Typical read length for each primer in base pairs (default: 800)
primer_length [Optional] Length of newly designed primers (default: 20)
min_gc [Optional] Minimum GC content for new primers (default: 0.4)
max_gc [Optional] Maximum GC content for new primers (default: 0.6)
min_tm [Optional] Minimum melting temperature in °C (default: 55.0)
max_tm [Optional] Maximum melting temperature in °C (default: 65.0)

Parameter Adaptation: Adjust parameters based on tissue quality, species, and condition. Literature values take priority, then official defaults, then tissue-specific adjustments.

What this file has done since we first saw it

Hashed on every crawl. A supply-chain change to an agent config is a question of when, not whether, so the history is kept rather than the latest state alone.

design_verification_primers is a skill published in the GitHub repository GGbond-bo/MemOmics-Agent (19 stars, last pushed 2d ago), licensed MIT. It adds 45 tokens to every session and 1,375 once invoked, about $0.0002 per session on Opus 5. A static security scan graded it A with 0 findings. No closer match exists in the catalogue, so it is treated as the original; first seen 2026-09-03.
