The cover letter is your manuscript's first impression on the editor. It is a persuasive document that must convince the editor to send your paper for peer review — not a restatement of your abstract.
What Editors Look For
Editors read cover letters to answer three questions:
- Is this paper in scope? — Does it fit the journal's mission and readership?
- Is it novel and significant? — Does it advance the field meaningfully?
- Is it ethically sound? — Are there conflicts of interest, prior presentations, or related submissions?
A good cover letter addresses all three in under one page.
The Universal 5-Paragraph Model
Paragraph 1: Hook + Identification
Purpose: Capture attention and identify the manuscript.
Elements:
- Journal name (proves you've targeted the submission)
- Manuscript type (research article, review, case report)
- Title
- Why this topic matters now (the hook)
Example:
Dear Dr. Smith,
We are pleased to submit our manuscript, "Gut Microbiome Alterations in Early-Stage Alzheimer's Disease: A Prospective Cohort Study," for consideration as a Research Article in Nature Medicine. With 50 million people affected by dementia worldwide and no disease-modifying treatments available, understanding modifiable risk factors has never been more urgent.
Paragraph 2: Novelty and Significance
Purpose: Demonstrate why your paper deserves peer review.
Elements:
- What gap does it fill?
- What is new compared to existing literature?
- What are the key findings?
- Why do they matter?
Do NOT repeat the abstract. Instead, narrate the significance.
Example:
Our study addresses a critical gap: while late-stage Alzheimer's pathology is well-characterised, the gut-brain axis in the preclinical phase remains poorly understood. In a cohort of 200 cognitively normal adults followed for 5 years, we identified specific microbial signatures that predicted conversion to mild cognitive impairment with 78% accuracy. These findings suggest that microbiome modulation could be a viable early intervention strategy — a hypothesis testable in clinical trials.
