Procedure
Phase 1 — Define the question precisely
Entry: the signal stated.
- Record the event as it will be counted: the term, the grading, and whether the analysis counts subjects or events.
- Record the exposure metric proposed and why it suits the event's mechanism. A delayed or cumulative toxicity analysed against peak concentration will show no relationship whether or not one exists.
- Record the time window: exposure up to the event, or total exposure. For events that end exposure, total exposure is confounded by survival — patients who tolerated longer have more of it.
