Who this is for

A clinical pharmacologist asked what exposure has to say about a safety signal — for a safety committee, a signal review, or a benefit–risk discussion where the question is whether exposure explains anything.

When to use this skill

  • Preparing the exposure contribution to a safety signal review.
  • Assembling exposure evidence for a periodic safety discussion.
  • Answering whether an observed event tracks exposure.

Procedure

Phase 1 — Define the question precisely

Entry: the signal stated.

  1. Record the event as it will be counted: the term, the grading, and whether the analysis counts subjects or events.
  2. Record the exposure metric proposed and why it suits the event's mechanism. A delayed or cumulative toxicity analysed against peak concentration will show no relationship whether or not one exists.
  3. Record the time window: exposure up to the event, or total exposure. For events that end exposure, total exposure is confounded by survival — patients who tolerated longer have more of it.
