Who this is for

A clinical pharmacologist or pharmacometrician checking the data-handling rules that determine what an analysis dataset contains — before the analysis runs, or afterwards to establish what was actually applied.

When to use this skill

Procedure

Phase 1 — BLQ handling

Entry: analysis plan located.

  1. Record the specified BLQ method and whether it is named specifically enough to reproduce — discard, substitute at zero, substitute at half the limit, or model the likelihood of censoring.
  2. Record the lower limit of quantification used and its source. A limit quoted in the plan that differs from the bioanalytical report is a finding, and it silently changes which records are censored.
  3. Record whether the method differs before and after the first quantifiable concentration, which is common and frequently unstated.
  4. Record the proportion of observations below the limit. The handling method matters in proportion to that number — a choice among methods is near-irrelevant at 2% and decisive at 40%, and a plan that specifies a method without reporting the proportion has not established whether the choice matters.
  5. Flag substitution methods applied where the censored proportion is high enough to bias parameter estimates.
